Medical faculty of Sultan Agung Islamic University was established on October 10 .. 1963, approximately 17 months after the foundation of Sultan Agung Islamic University on 20 May 1962 and then received SK 74/BST/P/64 listed No. 28 February 1964, signed by Moh. Sa'id official Bureau Chief of Private Higher Education Department of Higher Education and Science.

Physical building is located at Jalan Raya Semarang Kaligawe Km.4. Since its inception the Faculty of Medicine Unissula already have teaching hospitals in the same shade YBWSA (Yayasan Sultan Waqf Board) of Sultan Agung Islamic Hospital and its location adjacent to the Faculty of Medicine.
